Overview

Talquetamab + lenalidomide is a combination therapy under investigation for the treatment of multiple myeloma. Talquetamab is a bispecific monoclonal antibody (marketed as Talvey) that targets both G protein–coupled receptor, class C, group 5, member D (GPRC5D) on myeloma cells and CD3 antigen on T cells. By binding to these targets, it redirects and activates T cells to kill GPRC5D-expressing myeloma cells through cytotoxicity and cytokine release[2][4][6]. Lenalidomide (marketed as Revlimid) is an immunomodulatory small molecule that enhances immune system activity against cancer cells and has direct anti-tumor effects[3][8]. The combination leverages the immune-stimulating properties of lenalidomide with the targeted T-cell redirection of talquetamab for enhanced anti-myeloma efficacy[1].
