Overview

Talquetamab + PD-1 inhibitor is an investigational combination therapy being studied primarily for use in relapsed or refractory multiple myeloma (RRMM). Talquetamab is a bispecific monoclonal antibody that targets G protein-coupled receptor family C group 5 member D (GPRC5D) on multiple myeloma cells and CD3 on T cells, inducing T-cell-mediated cytotoxicity against myeloma cells. The PD-1 inhibitor component blocks the programmed cell death protein 1 (PD-1) receptor on T cells, preventing tumor-induced T cell exhaustion and enhancing anti-tumor immune responses. The rationale for this combination is that talquetamab-driven T cell activation may be potentiated by relief of PD-1-mediated suppression, thereby improving anti-myeloma efficacy, particularly in populations with an exhausted or immunosuppressive microenvironment[2]. This combination is being evaluated in early-phase clinical trials (not yet approved for any indication).
