Overview

Tamibarotene + arsenic trioxide is a combination therapy investigated for the treatment of relapsed or refractory acute promyelocytic leukemia (APL). Tamibarotene (Am80) is a synthetic retinoid and a selective agonist of the retinoic acid receptors alpha (RARα) and beta (RARβ), designed to induce terminal differentiation of leukemic cells and overcome resistance to all-trans retinoic acid (ATRA). Arsenic trioxide (ATO) is an antineoplastic agent that induces apoptosis and promotes the degradation of the PML-RARα fusion protein. This combination was evaluated in a Phase I clinical trial led by Northwestern University's Robert H. Lurie Comprehensive Cancer Center, with tamibarotene provided by CytRx Corporation, to determine the safety and efficacy of the regimen in patients who had relapsed after standard frontline therapy.
