Tamibarotene + daratumumab is an investigational **combination therapy** comprising tamibarotene, a synthetic oral selective retinoic acid receptor alpha (RARα) agonist, and daratumumab, a human monoclonal antibody targeting CD38. **Tamibarotene** promotes differentiation and inhibits proliferation in hematologic malignancies, particularly in cells with RARA overexpression, through selective activation of RARα. **Daratumumab** binds to CD38 on hematologic cancer cells, inducing cell death by direct apoptosis and multiple Fc-dependent immune mechanisms such as antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC), as well as immunomodulation by depleting CD38-positive immunosuppressive cells[3][6][8]. While both drugs have been combined with other agents (e.g., azacitidine, venetoclax for tamibarotene; bortezomib, lenalidomide, dexamethasone for daratumumab), specific clinical evidence for the tamibarotene + daratumumab combination is limited, and it would be experimental if pursued.
