Overview

ONO-7475 (tamnorzatinib) is an orally available small molecule inhibitor targeting the receptor tyrosine kinases Axl and Mer. It blocks Axl- and Mer-mediated signaling pathways involved in tumor cell proliferation, survival, invasion, angiogenesis, metastasis, and drug resistance. Overexpression of these kinases is associated with poor prognosis in several cancers[2][3][5]. ONO-4538 is the code name for nivolumab (Opdivo), a fully human monoclonal antibody that targets programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to enhance anti-tumor immune responses[6]. The combination of ONO-7475 and ONO-4538 is being investigated in early-phase clinical trials for safety and tolerability in patients with pancreatic cancer when used alongside standard-of-care chemotherapy (gemcitabine plus nab-paclitaxel)[1]. Both drugs are developed by Ono Pharmaceutical; nivolumab was co-developed with Bristol Myers Squibb outside Japan/Korea/Taiwan.
