Tamoxifen + tumor necrosis factor alpha is an experimental drug combination consisting of the selective estrogen receptor modulator (SERM) tamoxifen and the cytokine tumor necrosis factor alpha (TNF-alpha). Tamoxifen acts primarily as an antagonist of estrogen receptors in breast tissue and is widely used in hormone receptor-positive breast cancer. TNF-alpha is a pro-inflammatory cytokine with direct anti-tumor effects but also complex roles in immune regulation and cell death. Preclinical studies have shown that this combination can produce additive inhibitory effects on proliferation and induce apoptosis in certain cancer cell lines, such as human glioblastoma cells. The mechanism does not require estrogen/progesterone receptors or alteration of external TNF-alpha receptors; instead, it appears to involve enhanced induction of apoptosis when both agents are used together[1]. This combination remains investigational and has not been approved for clinical use.
