Overview

Tanamilast + cyclosporine is a combination therapy comprising the potent, selective phosphodiesterase 4 (PDE4) inhibitor tanamilast (also known as CHF 6001) and the calcineurin inhibitor cyclosporine. Tanamilast was developed by Chiesi Farmaceutici as a next-generation PDE4 inhibitor with high topical potency and low systemic exposure, primarily targeted at respiratory conditions like COPD and asthma, as well as ophthalmic diseases like dry eye. Cyclosporine is a widely used immunosuppressant that acts by binding to cyclophilin and inhibiting the phosphatase activity of calcineurin, thereby preventing the nuclear translocation of NFAT and subsequent T-cell activation. The combination of these two agents is explored for its ability to synergistically inhibit inflammatory pathways, particularly the production of cytokines like TNF-alpha and IL-2, offering a potential therapeutic strategy for chronic inflammatory disorders where monotherapy is insufficient.
