Tandutinib + cytarabine + daunorubicin is an experimental combination regimen mainly studied for the treatment of acute myeloid leukemia (AML), especially in patients with FLT3 internal tandem duplication (ITD)-positive disease. Tandutinib is an oral, small-molecule inhibitor of the receptor tyrosine kinase FLT3, which is often mutated in AML. Cytarabine is a nucleoside analog that interferes with DNA synthesis, and daunorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II. Preclinical and clinical data indicate that tandem administration of tandutinib synergizes with cytarabine and daunorubicin to produce enhanced antileukemic effects, particularly against FLT3 ITD-positive blasts, without increasing key chemotherapy-associated toxicities. The combination aims to improve overall response rates and may allow reduction of chemotherapy dosing while maintaining efficacy, potentially benefiting elderly or comorbid patients with AML[4][5][7].
