Tanespimycin + trastuzumab is a combination therapy consisting of tanespimycin (17-AAG, KOS-953), a heat shock protein 90 (HSP90) inhibitor, and trastuzumab (Herceptin), a monoclonal antibody targeting HER2. This combination was developed for HER2-positive metastatic breast cancer patients whose disease progressed during prior trastuzumab treatment. Tanespimycin inhibits HSP90, leading to degradation of client proteins including HER2, potentially resensitizing trastuzumab-resistant tumors to trastuzumab therapy.
