**Tanezumab + nonsteroidal anti-inflammatory drug** is a combination therapy consisting of tanezumab, a recombinant humanized monoclonal IgG2 antibody targeting nerve growth factor (NGF), administered together with a nonsteroidal anti-inflammatory drug (NSAID), such as naproxen or celecoxib. Tanezumab inhibits binding of NGF to its high-affinity receptor TrkA and low-affinity receptor p75, interrupting pain signaling pathways. NSAIDs reduce inflammation and pain primarily through inhibition of cyclooxygenase (COX) enzymes. The combination has been investigated for enhanced symptomatic relief in osteoarthritis (OA) and chronic pain, but studies show only modest improvement over tanezumab monotherapy while increasing risk of adverse events, especially rapidly progressive OA and joint replacements[1][3][5][7].
