The TARP peptide vaccine is a cancer immunotherapy developed by the National Cancer Institute (NCI) targeting the T-cell receptor alternate reading frame protein (TARP). TARP is a 58-amino acid protein that is highly over-expressed in approximately 95% of prostate cancer and 50% of breast cancer specimens. The vaccine typically consists of two investigational peptide agents: the native TARP 27-35 peptide (NSC 740703) and an epitope-enhanced TARP 29-37-9V peptide (NSC 740704). The epitope-enhanced version is modified to increase HLA-A*0201 binding affinity, thereby improving the stimulation of cytotoxic T-lymphocyte responses. In clinical trials for Stage D0 prostate cancer, the vaccine has been evaluated as a peptide emulsion with adjuvants (Montanide ISA 51VG and GM-CSF) or as an autologous peptide-pulsed dendritic cell vaccine, with the goal of slowing the rate of PSA rise and tumor growth.
