Tazarotene + minocycline is a combination therapy used primarily for the treatment of moderate to severe acne vulgaris. Tazarotene is a topical retinoid that, after conversion to its active metabolite tazarotenic acid, binds selectively to retinoic acid receptors (RARs), particularly RAR-beta and RAR-gamma, modulating gene expression involved in cell proliferation and inflammation[1]. Minocycline is an oral tetracycline-class antibiotic that inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, thereby reducing Cutibacterium acnes and other skin flora implicated in acne pathogenesis[6]. Clinical studies have shown that initial combination therapy with topical tazarotene and oral minocycline leads to significant improvement in both inflammatory and noninflammatory acne lesions. Maintenance regimens using either agent alone or in combination can sustain these improvements; however, long-term use of topical retinoid monotherapy may be preferred to minimize antibiotic exposure[2][3][4].
