Overview

TBIO-6517 + pembrolizumab is a combination investigational therapy for advanced solid tumors. TBIO-6517 is an oncolytic vaccinia virus, genetically attenuated to selectively infect and replicate in tumor cells. It is designed to stimulate local and systemic antitumor immune responses through direct oncolysis and immune activation. Pembrolizumab (Keytruda) is a monoclonal antibody checkpoint inhibitor that blocks the PD-1 receptor, lifting immune inhibition and promoting T-cell mediated cancer cell killing. This combination seeks to enhance antitumor immunity by pairing the immunogenic effects of intratumoral virotherapy (TBIO-6517) with systemic immune checkpoint blockade (pembrolizumab). Both direct intratumoral injection and intravenous modalities are tested.
