TCRαβ+ T cell-depleted hematopoietic stem cells are an ex vivo graft engineering technique used in haplo-hematopoietic stem cell transplantation. This method aims to effectively reduce the risk of graft-versus-host disease (GVHD) by selectively depleting TCRαβ+ T cells from the stem cell graft. The process typically involves magnetic sorting using devices like the CliniMACS® Plus, which allows for thorough depletion of TCRαβ+ T cells while maintaining high yields of CD34+ cells, TCRγδ+ T cells, and NK cells. This approach enables the infusion of mega-dose CD34+ cells, even in higher-weight patients, and has shown high primary engraftment rates.
