Teclistamab + daratumumab (often referred to as "Tec-Dara") is a combination therapy under investigation and in use for the treatment of multiple myeloma, particularly in relapsed/refractory or newly diagnosed settings. Teclistamab is a bispecific monoclonal antibody that targets B-cell maturation antigen (BCMA) on myeloma cells and CD3 on T-cells, redirecting T-cells to kill BCMA-expressing malignant plasma cells. Daratumumab is a monoclonal antibody targeting CD38, another surface protein highly expressed on myeloma cells, leading to direct cytotoxicity and immune-mediated cell death. The combination leverages complementary mechanisms—teclistamab’s T-cell engagement and daratumumab’s immunomodulatory effects—to enhance anti-myeloma activity. This regimen has shown promising efficacy as induction and maintenance therapy after autologous stem cell transplant in multiple myeloma patients[1][4][5][7][8].
