**Telmisartan + amlodipine** is an oral fixed-dose antihypertensive combination containing telmisartan, an angiotensin II receptor blocker, and amlodipine, a dihydropyridine calcium-channel blocker. It is used to treat hypertension, including as initial therapy for patients likely to require more than one antihypertensive medicine and as replacement or add-on therapy when blood pressure is inadequately controlled with either component alone. Telmisartan reduces angiotensin II-mediated vasoconstriction and aldosterone signaling through angiotensin II receptor type 1 blockade, while amlodipine relaxes vascular smooth muscle by inhibiting L-type calcium-channel-mediated calcium influx. The originator fixed-dose product is marketed by Boehringer Ingelheim as Twynsta.
