TOL is an investigational **combination chemotherapy regimen** comprising temozolomide, lomustine, and vincristine. Temozolomide and lomustine are DNA-alkylating cytotoxic agents, while vincristine disrupts mitotic microtubule formation. The Finnish Melanoma Group evaluated TOL with subcutaneous interferon-alpha, with or without vemurafenib, in the completed COBRA study for previously untreated unresectable advanced melanoma; interferon-alpha and vemurafenib are not components of TOL itself.
