Temsirolimus + radiation therapy is a combination treatment regimen investigated for the treatment of locally advanced non-small cell lung cancer (NSCLC). Temsirolimus (Torisel, CCI-779) is a small molecule, intravenous prodrug of sirolimus that functions as a selective inhibitor of the mammalian target of rapamycin (mTOR), a serine/threonine kinase that plays a central role in regulating cell proliferation, apoptosis, and angiogenesis. By inhibiting the mTORC1 complex, temsirolimus disrupts the PI3K/AKT/mTOR signaling pathway, which is frequently dysregulated in lung cancer. In clinical studies conducted by Washington University School of Medicine, temsirolimus was administered concurrently with thoracic radiation therapy to evaluate the maximum tolerated dose and safety profile, aiming to exploit the potential radiosensitizing effects of mTOR inhibition.
