This is a combination of three antiviral agents: - **Tenofovir alafenamide** is a nucleotide reverse transcriptase inhibitor (NRTI) that acts as a prodrug of tenofovir, inhibiting HIV-1 and hepatitis B virus polymerases by causing DNA chain termination after incorporation into viral DNA[8][10]. - **Ledipasvir** is an inhibitor of the hepatitis C virus (HCV) NS5A protein, which is essential for viral replication and assembly[2][3]. - **Sofosbuvir** is a nucleotide analog inhibitor targeting the HCV NS5B RNA-dependent RNA polymerase, leading to chain termination during viral RNA replication[2][3]. This combination would theoretically provide broad-spectrum antiviral activity against both HIV and HCV. However, there are no approved fixed-dose products containing all three agents together; they are sometimes coadministered in clinical practice for patients coinfected with HIV and HCV. The safety profile has been studied in such settings, showing no clinically significant pharmacokinetic interactions requiring dose adjustment when used together[1][5]. All three drugs are developed by Gilead Sciences.
