Overview

TER-2013 + fulvestrant is an **experimental combination therapy** under clinical investigation for **advanced solid tumors with AKT/PI3K/PTEN pathway alterations**, including certain types of breast, lung, ovarian, endometrial, esophageal, cervical, and head or neck cancers. TER-2013 is being studied as both a monotherapy and in combination with fulvestrant. Fulvestrant is a selective estrogen receptor degrader (SERD), functioning by antagonizing the estrogen receptor and promoting its degradation, thus inhibiting estrogen-driven tumor growth. The rationale for the combination is to co-target the estrogen signaling axis (via fulvestrant) and the PI3K/AKT pathway (via TER-2013), which are both implicated in endocrine resistance and tumor survival in hormone receptor-positive malignancies. The combination is currently in Phase 1/2 clinical trials to establish safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity[2][5].
