Overview

TERN-101 + TERN-501 is an investigational oral combination therapy under development for the treatment of non-alcoholic steatohepatitis (NASH). The combination consists of two small-molecule agents: - **TERN-101**, a potent and highly selective farnesoid X receptor (FXR) agonist, which modulates bile acid metabolism and has anti-inflammatory and anti-fibrotic effects in the liver. - **TERN-501**, a selective thyroid hormone receptor beta (THRβ) agonist, which reduces liver fat content by promoting lipid metabolism in hepatocytes. Preclinical and clinical studies have shown that combining these two mechanisms may provide additive or synergistic benefits for NASH, improving both steatosis and fibrosis more than either agent alone. The combination has demonstrated efficacy in reducing liver fat content, improving noninvasive biomarkers of fibro-inflammation, and favorably impacting lipid profiles with a safety profile similar to monotherapy arms[2][3][8]. Both drugs are being developed by Terns Pharmaceuticals.
