Testosterone undecanoate + letrozole is a combination of two pharmaceutical agents with distinct mechanisms of action. Testosterone undecanoate is a long-acting ester of testosterone used primarily in the treatment of male hypogonadism and as androgen therapy in transgender men. It acts as an agonist at the androgen receptor to promote the development and maintenance of male secondary sexual characteristics and support bone and muscle health[2][3][7]. Letrozole is a third-generation non-steroidal aromatase inhibitor indicated for estrogen-dependent breast cancers in postmenopausal women. It works by competitively inhibiting aromatase (CYP19A1), thereby reducing estrogen synthesis from androgen precursors[5][6]. The combination may be considered experimentally or off-label to manage hormone levels—such as suppressing estrogenic side effects during testosterone therapy or for specific research protocols—but it is not an approved fixed-dose combination.
