Overview

**tetracycline + metformin** is a combination of two drugs: tetracycline (a broad-spectrum antibiotic) and metformin (an oral antidiabetic agent). This combination is primarily under investigation as a therapeutic approach to restore tetracycline's efficacy against multidrug-resistant bacterial infections, by utilizing metformin as an adjuvant. Metformin potentiates tetracycline (especially doxycycline) activity by increasing bacterial membrane permeability, dissipating membrane potential, promoting intracellular accumulation of tetracycline, and undermining the function of the proton motive force-driven efflux pump, thus overcoming bacterial resistance. Additionally, metformin modulates the host immune response and reduces inflammatory responses. The primary target indications are resistant bacterial infections, and preclinical and early-phase clinical studies support its promise in overcoming antibiotic resistance[1][2][3][4][5].
