Drug intelligence / Profile preview
TG01 + QS-21 is an investigational therapeutic cancer vaccine combination consisting of TG01, a polyvalent peptide vaccine, and the saponin-based adjuvant QS-21. TG01 is composed of seven synthetic long peptides (SLPs) that target the most frequent mutations in the KRAS oncogene at codons 12 and 13, specifically G12D, G12V, G12R, G12C, G12S, G12A, and G13D. The vaccine is designed to stimulate a robust, mutation-specific T-cell response (both CD4+ and CD8+) against tumor cells harboring these RAS mutations. QS-21, derived from the *Quillaja saponaria* tree, is a potent adjuvant that enhances the immunogenicity of the peptides by promoting both cellular and humoral immune responses. Originally developed by Targovax (now Circio) for gastrointestinal cancers, this combination is currently being evaluated in clinical trials sponsored by Oslo University Hospital for patients with RAS-mutated multiple myeloma and smoldering multiple myeloma.
