Overview

TG6002 + flucytosine is an investigational combination therapy consisting of TG6002, a genetically engineered oncolytic vaccinia virus, administered together with the prodrug flucytosine (5-FC). TG6002 is designed to selectively replicate in and lyse tumor cells, inducing immunogenic cell death. It has been engineered to express the chimeric yeast FCU1 gene, enabling localized conversion of flucytosine into 5-fluorouracil (5-FU), a potent cytotoxic agent, within the tumor microenvironment. This strategy aims to maximize anti-tumor efficacy through the combination of direct oncolysis, immune activation, and local chemotherapy, while minimizing systemic toxicity. The therapy is being developed primarily for solid tumors, particularly colorectal and gastrointestinal adenocarcinoma, as well as glioblastoma.
