TGRN-321 + envafolimab is an investigational combination therapy being developed by Simcere Pharmaceutical (Jiangsu Xiansheng) in collaboration with Tagworks Pharmaceuticals. TGRN-321 is a first-in-class antibody-drug conjugate (ADC) targeting Tumor-Associated Glycoprotein 72 (TAG72), utilizing Tagworks' proprietary 'Click-to-Release' technology. This platform allows for the systemic administration of a prodrug that is selectively activated at the tumor site by a secondary 'trigger' molecule, potentially enhancing the therapeutic index by reducing off-target toxicity. Envafolimab (KN035) is a subcutaneous PD-L1-targeting single-domain antibody (nanobody) already approved in China for certain oncology indications. The combination aims to leverage the targeted cytotoxic effects of the TAG72 ADC with the immune-potentiating effects of PD-L1 checkpoint inhibition to treat various TAG72-expressing solid tumors.
