Thalidomide + dexamethasone is an oral combination regimen used primarily for the treatment of newly diagnosed multiple myeloma. Thalidomide is an immunomodulatory agent with anti-inflammatory and anti-cancer properties, acting through mechanisms such as inhibition of angiogenesis, modulation of cytokine production, and enhancement of natural killer cell activity. Dexamethasone is a synthetic glucocorticoid corticosteroid that exerts potent anti-inflammatory and immunosuppressive effects. The combination has demonstrated superior response rates compared to dexamethasone alone in clinical trials but carries increased risks for adverse events such as deep vein thrombosis and peripheral neuropathy[1][2][5][6][8].
