Overview

This is an oral combination chemotherapy regimen consisting of thalidomide (an immunomodulatory agent), dexamethasone (a corticosteroid), and cyclophosphamide (an alkylating agent). It is primarily used for the treatment of multiple myeloma, both as initial therapy and in relapsed/refractory settings. Cyclophosphamide works by cross-linking DNA strands to inhibit cell division in rapidly dividing cells. Thalidomide acts as a cancer growth blocker by inhibiting angiogenesis (formation of new blood vessels) required for tumor growth and modulating the immune system to enhance anti-myeloma activity. Dexamethasone reduces inflammation and enhances the efficacy of chemotherapy agents[1][2][3][6]. The combination is typically administered orally in cycles and has demonstrated effectiveness with manageable toxicity profiles compared to some other regimens.
