This is a combination of two drugs, thalidomide and etanercept. Thalidomide is an immunomodulatory small molecule with anti-inflammatory, anti-angiogenic, and immunosuppressive properties. It acts primarily by inhibiting tumor necrosis factor alpha (TNF-α) production and modulating other cytokines. Etanercept is a biologic fusion protein that functions as a tumor necrosis factor inhibitor; it consists of the extracellular ligand-binding portion of the human TNF receptor linked to the Fc portion of human IgG1, binding both TNF-α and TNF-β to block their pro-inflammatory effects. This combination would theoretically provide synergistic inhibition of TNF-mediated inflammation through different mechanisms—thalidomide reducing production/secretion and etanercept neutralizing circulating cytokine—but there are no approved indications or established clinical uses for this specific drug combination.
