Thalidomide + interferon is a combination therapy consisting of thalidomide, an oral immunomodulatory and anti-angiogenic small molecule, and interferon (typically recombinant human interferon-alpha), a cytokine with antiviral, antiproliferative, and immunomodulatory properties. Thalidomide acts by modulating the immune system, inhibiting tumor necrosis factor-alpha (TNF-α) production, and suppressing angiogenesis. Interferons bind to cell surface receptors to activate intracellular signaling pathways that enhance immune responses against malignant cells. This combination has been investigated for the treatment of recurrent or refractory hematologic malignancies such as acute myeloid leukemia (AML) and multiple myeloma. The rationale for combining these agents is their complementary mechanisms—thalidomide’s immunomodulation/anti-angiogenesis with interferon's direct stimulation of antitumor immunity[3][4][7].
