Overview

This investigational combination immunotherapy, developed by the Mayo Clinic, consists of autologous dendritic cells and the pneumococcal 13-valent conjugate vaccine (Prevnar 13). It is being evaluated for the treatment of unresectable primary liver cancers, specifically hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (iCCA). The treatment strategy involves harvesting a patient's white blood cells to generate dendritic cells, which are then cultured in a laboratory and injected intratumorally to stimulate an immune response against tumor antigens. Prevnar 13 is administered intramuscularly as an adjuvant to further boost the immune system's activity. This regimen is typically applied following high-dose external beam radiotherapy and is often combined with immune checkpoint inhibitors like atezolizumab and bevacizumab to overcome tumor-mediated immunosuppression and enhance anti-tumor immune responses.
