Overview

Therapeutic autologous lymphocytes (Barbara Ann Karmanos Cancer Institute) refers to an investigational adoptive cell therapy protocol developed for the treatment of resected stage III or IV melanoma. The approach involves a multi-step immunotherapy regimen: patients are first sensitized with an autologous tumor cell vaccine (composed of irradiated tumor cells) and sargramostim (GM-CSF) to stimulate dendritic cell maturation and antigen presentation. Subsequently, autologous T lymphocytes are harvested, activated ex vivo, and re-infused into the patient. This cellular transfer is supported by the administration of aldesleukin (IL-2) to promote the in vivo expansion and survival of the transferred T cells. The mechanism of action relies on the generation and expansion of tumor-specific T cells that can recognize and lyse melanoma cells expressing specific cancer antigens. Clinical evaluation was primarily conducted through Phase II trials at the Karmanos Cancer Institute.
