Overview

Thiazide + atenolol is a fixed-dose combination therapy consisting of the thiazide diuretic hydrochlorothiazide (12.5 mg) and the cardioselective beta-blocker atenolol (50 mg). This specific combination was evaluated as a treatment arm in The Indian Polycap Study (TIPS), a Phase 3 clinical trial led by St. John's Research Institute and sponsored by Cadila Pharmaceuticals. The combination aims to reduce cardiovascular risk by lowering blood pressure through two distinct mechanisms: hydrochlorothiazide inhibits the sodium-chloride symporter in the distal convoluted tubule of the kidney to promote diuresis and reduce blood volume, while atenolol selectively antagonizes beta-1 adrenergic receptors to reduce heart rate and cardiac contractility. It was studied in middle-aged and elderly individuals with at least one cardiovascular risk factor as part of a larger evaluation of polypill components.
