Overview

thioredoxin-L2(20–38)3 is an experimental pan-Human Papillomavirus (HPV) vaccine candidate designed to provide broad protection against a wide range of HPV genotypes. The construct is a recombinant fusion protein consisting of a thioredoxin scaffold, typically derived from the hyperthermophilic archaeon *Pyrococcus furiosus* (PfTrx), which serves as a stable carrier for three tandemly repeated copies of a highly conserved epitope from the HPV L2 minor capsid protein (amino acids 20–38). Unlike current L1-based vaccines (e.g., Gardasil) that are type-specific, this L2-based approach targets a conserved region to induce cross-neutralizing antibodies effective against both high-risk (oncogenic) and low-risk (wart-causing) HPV types. The vaccine is primarily being developed through research collaborations involving the German Cancer Research Center (DKFZ) and Johns Hopkins University.
