Overview

Thymidine‑5'‑thiophosphate is a synthetic nucleotide analog in which the phosphate group at the 5' position of thymidine is replaced by a thiophosphate group. It belongs to the class of pyrimidine 2'-deoxyribonucleosides and has been studied as an experimental small molecule. Thymidine analogs are often used in biochemical research to probe DNA synthesis and repair mechanisms or as potential inhibitors of viral polymerases. Recent computational studies have identified thymidine‑5'‑thiophosphate as a potential inhibitor of SARS-CoV‑2 RNA-dependent RNA polymerase through molecular docking and dynamics simulations; however, there are no approved clinical uses for this compound[1][10][8]. Its mechanism may involve competitive inhibition at nucleic acid processing enzymes.
