Overview

KHK2866 (tiamilimab) is a humanized IgG1 monoclonal antibody that acts as an agonist of the OX40 (CD134) receptor, a co-stimulatory molecule primarily expressed on activated T cells. By binding to OX40, tiamilimab enhances T-cell activation, proliferation, and survival, thereby promoting a robust anti-tumor immune response. Paclitaxel is a well-established taxane chemotherapy agent that stabilizes microtubules, leading to mitotic arrest and subsequent apoptosis of rapidly dividing tumor cells. The combination of tiamilimab and paclitaxel is being investigated for potential synergistic effects in advanced solid tumors, where paclitaxel-induced immunogenic cell death may release tumor antigens and create an inflammatory microenvironment that facilitates the immune-stimulatory activity of the OX40 agonist.
