Overview

Tifcemalimab + toripalimab is a combination of two monoclonal antibodies used in cancer immunotherapy. Tifcemalimab is a humanized IgG4 monoclonal antibody targeting B and T lymphocyte attenuator (BTLA), an inhibitory co-signaling receptor expressed on B cells, T cells, and NK cells. It blocks the interaction between BTLA and its ligand HVEM. Toripalimab is a monoclonal antibody that targets programmed death protein-1 (PD-1), blocking the PD-1 pathway to enhance anti-tumor immune responses. The combination aims to synergistically improve antigen-specific anti-tumor T cell responses by dual checkpoint inhibition. This regimen has shown promising efficacy and manageable safety in early-phase clinical trials for extensive-stage small cell lung cancer (ES-SCLC), limited-stage SCLC (LS-SCLC), lymphoma, and esophageal squamous cell carcinoma[1][2][3][4][5][6].
