Tiragolumab + atezolizumab + cisplatin + etoposide is a four-drug combination therapy studied primarily for **extensive-stage small-cell lung cancer (ES-SCLC)**. Tiragolumab is a monoclonal antibody that targets TIGIT (T-cell immunoreceptor with Ig and ITIM domains), functioning as an immune checkpoint inhibitor. Atezolizumab is a monoclonal antibody against PD-L1 (programmed death-ligand 1), also functioning as an immune checkpoint inhibitor. Cisplatin and etoposide are established cytotoxic chemotherapies. The rationale for combining tiragolumab and atezolizumab is to enhance the immune-mediated anti-tumor response by dual checkpoint inhibition, while cisplatin and etoposide provide direct cytotoxic activity against tumor cells. This regimen has been evaluated and well tolerated in clinical trials, with the addition of tiragolumab showing no additional efficacy benefit over atezolizumab plus chemotherapy in ES-SCLC but still under further study for other cancer types[2][4][1].
