Overview

This is an investigational combination therapy consisting of three monoclonal antibodies: **tiragolumab** (an anti-TIGIT antibody), **atezolizumab** (an anti–PD-L1 antibody), and **daratumumab** (an anti-CD38 antibody). The combination is being studied primarily in relapsed/refractory multiple myeloma and potentially other advanced solid tumors. Tiragolumab blocks TIGIT, enhancing T-cell and NK cell immune responses; atezolizumab blocks PD-L1, preventing inhibition of T-cell activity by tumor cells; daratumumab targets CD38, inducing immunologic cell death in plasma cells. The rationale is to synergistically target multiple immune checkpoints and tumor pathways to improve antitumor efficacy in difficult-to-treat cancers.
