This is a four-drug combination regimen consisting of the anti-PD-1 monoclonal antibody tislelizumab, the microtubule inhibitor albumin-bound paclitaxel (nab-paclitaxel), the platinum-based DNA crosslinker cisplatin, and the antimetabolite 5-fluorouracil. Tislelizumab blocks PD-L1/PD-L2-mediated signaling to restore T-cell function and promote immune-mediated tumor cell killing. Albumin-bound paclitaxel disrupts microtubule dynamics, inhibiting mitosis in rapidly dividing cells. Cisplatin forms DNA crosslinks that prevent replication and transcription, leading to apoptosis in cancer cells. 5-Fluorouracil inhibits thymidylate synthase and incorporates into RNA/DNA, disrupting nucleic acid synthesis and function. This combination is being investigated primarily as neoadjuvant or first-line therapy for locally advanced or metastatic esophageal squamous cell carcinoma (ESCC) and other solid tumors[3][9][7].
