Overview

This combination therapy consists of the anti-PD-1 monoclonal antibody tislelizumab, the retinoid all-trans retinoic acid (ATRA), and standard chemotherapy (typically paclitaxel combined with cisplatin or carboplatin). It is being investigated by Fudan University as a first-line treatment for locally advanced or metastatic esophageal squamous cell carcinoma (ESCC). The rationale behind adding ATRA is to modulate the immunosuppressive tumor microenvironment, specifically by targeting myeloid-derived suppressor cells (MDSCs), thereby potentially enhancing the anti-tumor activity of the PD-1 checkpoint inhibitor tislelizumab. Chemotherapy is included to provide direct cytotoxic effects and potentially induce immunogenic cell death, further priming the immune system.
