Overview

This combination therapy pairs tislelizumab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, with atorvastatin, an HMG-CoA reductase inhibitor. Tislelizumab functions as an immune checkpoint inhibitor, preventing PD-1 from binding to its ligands (PD-L1 and PD-L2) to restore T-cell mediated anti-tumor activity. Atorvastatin, traditionally used for lipid-lowering, is investigated here for its potential pleiotropic effects, including the modulation of the tumor microenvironment and enhancement of immunotherapy efficacy. The combination is primarily being explored by the Army Specialty Medical Center (Daping Hospital) for the treatment of advanced non-small cell lung cancer (NSCLC), based on the hypothesis that statins may overcome resistance to PD-1 blockade.
