Overview

This is a combination investigational oncology regimen consisting of **tislelizumab**, a humanized IgG4 anti-programmed cell death protein-1 (PD-1) monoclonal antibody; **BGB-15025**, a selective small-molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1); and chemotherapy. Tislelizumab blocks the PD-1/programmed death-ligand 1 (PD-L1) immune checkpoint, which releases T-cell inhibition and potentiates immune-mediated tumor cell killing. BGB-15025 inhibits HPK1, a negative regulator of T-cell receptor signaling, to further enhance T-cell activation. Preclinical and clinical data suggest that combining HPK1 inhibition (BGB-15025) with PD-1 blockade (tislelizumab) and chemotherapy may yield synergistic anti-tumor effects. This combination is under clinical investigation in advanced solid tumors, including non-small cell lung cancer[1][3][4][5].
