This is a combination therapy comprised of three main components: **tislelizumab**, an anti–PD-1 monoclonal antibody; **LBL-007**, an anti–LAG-3 monoclonal antibody; and **chemotherapy** (typically gemcitabine and cisplatin in published studies). Tislelizumab blocks the PD-1 protein on T cells to enhance immune system attack on cancer cells. LBL-007 targets the lymphocyte activation gene-3 (LAG-3), a checkpoint that inhibits T-cell activation, and helps further augment antitumor immune responses. Chemotherapy adds cytotoxic effects and may enhance immunogenic cell death, further boosting the efficacy of the immuno-oncology agents. The combination is being investigated primarily in first-line, unresectable, locally advanced or metastatic esophageal squamous cell carcinoma (ESCC) and in advanced nasopharyngeal carcinoma (NPC), with studies reporting high objective response and disease control rates in these settings[1][2][3][4].
