Overview

This is a combination therapy consisting of three agents: tislelizumab, dasatinib, and quercetin. Tislelizumab is a humanized monoclonal antibody targeting programmed death receptor 1 (PD-1), developed to block the interaction between PD-1 and its ligands PD-L1/PD-L2, thereby restoring T-cell function and promoting immune-mediated antitumor activity[6][9]. Dasatinib is an FDA-approved small molecule tyrosine kinase inhibitor that targets multiple kinases involved in cancer cell proliferation and survival; it also exhibits pro-apoptotic effects on senescent cells[4][5]. Quercetin is a naturally occurring flavonoid with antioxidant, anti-inflammatory, and senolytic properties; it can induce apoptosis in certain types of senescent cells[7][10]. The combination of dasatinib plus quercetin has been studied as a "senolytic" regimen to selectively eliminate senescent cells implicated in aging-related diseases and tumor microenvironment modulation[2][4][5]. This triple combination has entered clinical trials as neoadjuvant therapy for resectable head and neck squamous cell carcinoma (HNSCC), aiming to reverse immunosenescence, enhance antitumor immunity, and improve treatment outcomes by combining immune checkpoint inhibition with targeted elimination of senescent cells[1][3].
