Overview

Tislelizumab + etoposide is a combination regimen consisting of tislelizumab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), and etoposide, a small-molecule topoisomerase II inhibitor. Developed by BeiGene (now BeOne Medicines), this combination is typically administered intravenously in conjunction with a platinum agent (cisplatin or carboplatin) for the first-line treatment of extensive-stage small cell lung cancer (ES-SCLC). In the phase 3 RATIONALE-312 trial, the addition of tislelizumab to etoposide and platinum chemotherapy demonstrated a statistically significant and clinically meaningful improvement in overall survival compared to placebo plus chemotherapy. The combination has been approved by China's National Medical Products Administration (NMPA) and recommended by the European Medicines Agency's CHMP for first-line ES-SCLC, and is also being investigated in phase 2 trials for limited-stage small cell lung cancer (LS-SCLC).
