This combination therapy consists of tislelizumab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), and Huaier granule, a traditional Chinese medicine (TCM) extract derived from the fungus *Trametes robiniophila Murr*. Currently being evaluated by Tongji Hospital as a first-line treatment for unresectable hepatocellular carcinoma (HCC), the regimen utilizes the synergistic potential of immune checkpoint inhibition and the multi-targeted anti-tumor properties of Huaier. Huaier granule contains bioactive components such as polysaccharides and proteins that inhibit tumor cell proliferation, induce apoptosis, and suppress angiogenesis. Furthermore, Huaier has been shown to modulate the tumor microenvironment by promoting CD8+ T cell infiltration and upregulating PD-L1 expression, which may enhance the therapeutic efficacy of PD-1 blockade in HCC patients.
