Tislelizumab + paclitaxel + carboplatin is a combination regimen used primarily as first-line therapy for advanced or metastatic squamous non-small-cell lung cancer (sq-NSCLC). Tislelizumab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to enhance anti-tumor immune responses. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division, while carboplatin is a platinum-based chemotherapeutic that induces DNA crosslinking and apoptosis in rapidly dividing cells. The combination leverages immunotherapy with cytotoxic chemotherapy to improve progression-free survival and overall response rates compared to chemotherapy alone[1][5][6]. This regimen has demonstrated superior clinical benefit and manageable safety profile in phase III trials for sq-NSCLC[1][5]. It has also been investigated in other cancers such as triple-negative breast cancer (TNBC)[2].
