This combination therapy consists of tislelizumab, a humanized IgG4 monoclonal antibody targeting the programmed cell death-1 (PD-1) receptor, and the cytotoxic chemotherapeutic agents paclitaxel and cisplatin. It is being investigated as a conversion therapy for patients with initially unresectable esophageal squamous cell carcinoma (ESCC). The goal of this regimen is to achieve tumor downstaging to allow for R0 surgical resection. Tislelizumab works by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor immune responses. Paclitaxel is a microtubule stabilizer that interferes with mitosis, while cisplatin is a platinum-based alkylating agent that causes DNA cross-linking, leading to apoptosis.
