**Tislelizumab + surzebiclimab** is an investigational combination of two monoclonal antibodies designed as a cancer immunotherapy. Tislelizumab is a humanized IgG4 monoclonal antibody targeting the programmed death-1 (PD-1) receptor, developed to inhibit the PD-1/PD-L1 pathway and restore T-cell activity, thereby enhancing immune-mediated tumor destruction. Surzebiclimab is an anti-TIM-3 monoclonal antibody targeting T-cell immunoglobulin and mucin domain-containing protein 3 (TIM-3), an immune checkpoint involved in T-cell exhaustion and resistance to PD-1 inhibition. The rationale for this combination is to simultaneously block two inhibitory pathways on T cells—PD-1 and TIM-3—to further enhance anti-tumor immune responses beyond PD-1 blockade alone. This combination is currently under clinical investigation, including studies focused on advanced solid tumors such as recurrent or metastatic head and neck squamous cell carcinoma. Both antibodies are products of advanced immunotherapy research aiming to overcome tumor immune resistance and promote durable responses in cancer patients[1][3][5][7].
